Pre-Eater of Worlds/Brain of Cthulhu

Defeating your first bosses vastly opens your options: After the Eye of Cthulhu is defeated, you should explore the Underground Jungle, fight the Blood Moon and Goblin Army, and challenge the newly unlocked Acid Rain event. Defeating Desert Scourge unlocks the Giant Clam miniboss and is the point where you should explore the Sunken Sea. Crabulon can also be defeated for weapons and a Rage Mode upgrade. Lastly, maxing out your health and mana will help you prepare for your first major step in progression.

After defeating the The Hive Mind or The Perforators, you can now mine Aerialite, found in Floating Islands. You can also fight Queen Bee and Deerclops to help prepare you for Skeletron.

Pre-Wall of Flesh

With Skeletron defeated, you should head for the Abyss and loot its newly unlocked Ancient Treasure Chests for gear that will help you defeat The Slime God, which drops useful materials and gear. Additionally, you can now use Blood Orbs at an Alchemy Table to make most available potions. When you are prepared, defeat the Wall of Flesh and enter hardmode.
