The Mushroom Plasma Root is a permanent power-up item obtained from the Treasure Bag dropped by Crabulon. Upon consumption, it increases the Rage Mode duration by one second. There are two other items that function identically to this: the Infernal Blood and the Red Lightning Container. It can only be used once.
